The Nucleic Acid of the Eggs of Arbacia Punctulata

چکیده
Quantitative determinations of the phosphorus distribution in the eggs of a variety of species by Tschernorutzki (l), Massing (2), and Needham and Needham (3) have indicated the probable presence of nucleoproteins in unfertilized ova. Unfortunately, however, the presence of vitellins in such cells renders the exact interpretation of such analyses somewhat difficult since, with current methods of analysis, it is possible that phosphoproteins of this type may be mistaken for nucleoproteins. In view of these facts it is of interest to determine the presence of nucleic acid in unfertilized ova by means of an actual isolation, which permits an examination of the characteristics of the substance isolated. For this purpose we have selected the eggs of the sea urchin, Arbaciu punctulata, whose alecithal nature facilitates the ease of the necessary chemical manipulations. In this connection it should be noted that Mathews (4) states: “The author got a substance with some of the properties of nucleic acid in some quantity from the unfertilized eggs of the sea urchin. It could not be positively identified, however, as the quantity was too small.”

Chemotaxis of Arbacia punctulata spermatozoa to resact, a peptide from the egg jelly layer
Resact, a peptide of known sequence isolated from the jelly layer of Arbacia punctulata eggs, is a potent chemoattractant for A. punctulata spermatozoa. The chemotactic response is concentration dependent, is abolished by pretreatment of the spermatozoa with resact, and shows an absolute requirement for millimolar external calcium. A. punctulata spermatozoa do not respond to speract, a peptide ...
متن کاملCharacterization of calmodulin and calmodulin isotypes from sea urchin gametes.
Procedures were developed for the purification of calmodulin from the eggs and from the sperm of the sea urchin Strongylocentrotus purpuratus and Arbacia punctulata. Two forms of calmodulin were isolated from A. punctulata eggs, one from each of the other three sources. All five calmodulins are similar to vertebrate calmodulin as judged by their activation of bovine brain cyclic nucleotide phos...
متن کاملOxidative Phosphorylation by a Cell-free Particulate System from Unfertilized Arbacia Eggs
1. A cell-free particulate system capable of effecting oxidative phosphorylation has been prepared from unfertilized eggs of Arbacia punctulata. A substantial increase in phosphorylation can be produced by addition of alpha-ketoglutarate, oxalacetate, or succinate, the magnitude of the increase being greatest with alpha-ketoglutarate. The activity of the phosphorylating system is sharply depend...
متن کاملIdentification of a sperm receptor on the surface of the eggs of the sea urchin Arbacia punctulata
The possibility that the surface of the egg of the sea urchin Arbacia punctulata contains a species-specific receptor for sperm has been investigated. The extent of fertilization of eggs of A. punctulata, which is proportional to the number of sperm, is unaffected by the presence of either eggs or membranes prepared from eggs of Strongylocentrotus purpuratus. In marked contrast, membranes prepa...
متن کاملA Note on the Respiration of Arbacia Eggs
Methods used in preparing Arbacia eggs for respiration studies, in carrying through the manometric determinations, and in estimating egg quantities have been reexamined. Discrepancies in previous results are almost entirely due to a steady error in measuring egg volume by centrifuging. Volumes so obtained averaged 80 per cent too high. The respiration of unfertilized eggs of Arbacia punctulata ...
